Check out http://www.andrewgrant.org/2008/03/30/how-to-sync-an-iphone-with-two-or-more-computers.html (are probably other URLs too if you Google but that was just one I had bookmarked that I tried with) - I did try this for a while but found it was pretty flakey and didn't work too well so just see how it works for you.

I faced the same issue for different reasons. I am planning to upgrade my desktop from Vista to Win 7 via clean install (not upgrade) over the coming weekend so needed to do something about my Itunes lib both media files and Iphone apps and also thought it'd be worthwhile to use my laptop for my Iphone/Itouch/Ipod sync as I use the laptop while travelling. My laptop has Win 7 on it.
On the apple site in the knowledge base section you can find comprehensive details on 2 options you can use; to backup and transfer via DVD or to back/transfer via your Iphone/ITouch/Ipod devices. Before you do any of the above you should know that you can authorize up to 5 PC's to share your lib and your Itunes account which is what I first did on my laptop.
Then I choose the first option and simply used the backup lib to DVD option on Itunes to create the DVD's and restored them on my laptop. To make it simpler load Itunes on your second PC (once you've auth it) and then insert the DVD it'll automatically recognize it and run the restore. I had 3 DVD's due to the size of my media/apps and all of them restored without any problems and then to test this I connected my old 1st gen ITouch which was recognized without any problems on my laptop. Going to be brave and connect my IPhone tonight :)
Hope the above helps. I am also going to try the backup restore via IPhone over the weekend. Finally, there is also a simple way to manually backup your entire media folder on Itunes and copy it to another PC and even merge it with new media files that you may have on the new PC.
