When plugging my headphones into my laptop, I can hear some interference and even when the sound is turned down completely, I still hear this humming end beeping noise.

It is quite irritating.

What could be causing this?
Wi-Fi?
Incorrectly assembled laptop?

Usually its the hard drive that would cause interference. The WiFi's frequency is too high to cause interference that you'll hear.

Your laptop's sound card is probably not well shielded.

See if the effect of the interference is reduced when you put the master volume on like 10% and the WAVE and other outputs on 100%

Where do I change these settings?

Open Volume Control/Mixer. Usually the Volume Control icon is in the system tray.

I'm not sure how you do this in Windows 7 though :(

check that line in is muted...

also, does this happen on battery or mains use? it shouldnt happen on battery use if it's a grounding problem...

I've unplugged it from mains and the sound is still there

So I've found what is causing the interference.

It's my optical USB mouse!

I have the Genius Xscroll.
Will buying a laser mouse solve the interference?

I solved the interference problem by plugging the mouse into another USB/eSATA combo port on the other side of the laptop :)
