No focus points

If you're taking a landscape shot - or evening shot of the city skyline - and there no focus point or its too dark to focus ... What's the best way?

Could setting a high F stop (ie 32 or higher) make everything in the frame clear? But then beef up ISO or tripod with higher exposure to compensate for light?
 
There's almost always something you can focus on, even if only manually.

Most lenses have a sweet spot somewhere in the mid-range aperture where they are sharpest. F/32 probably isnt it . . . and you'd risk seeing every little flack of dust on your sensor.
