TrueCrypt for a whole drive/partition is an excellent option. Though if you just want to protect some data like a file or folder, which may be the better option as locking the company or owner out of their PC could cost you your job or cause trouble at the least, use something like WinRAR and use "Store" for compression and encrypt file names and use a strong long password (UPPERCASE + lowercase + numbers + other characters such as &@#) and you can consider it safe.
I have to say though, whatever you do or store on a work PC should not be considered private, I'm all for privacy but storing your private "document" in someone else's "case" (without their permission) and having a fit when they look at it is silly.
