Update: Jigdo is really awesome, busy downloading my distro now. Only thing is that it only works with the "alternate" and "server". These are the ones I anyway want so not issue for me.

How do I trigger an update from the ISO?
Should I reboot with the disc in and boot from the Disc? or do the update from within Ubuntu?

Add the CD to your repos and change your version information.

To upgrade from Ubuntu 9.04 on a desktop system, press Alt+F2 and type in "update-manager -d" (without the quotes) into the command box. Update Manager should open up and tell you: New distribution release '9.10' is available. Click Upgrade and follow the on-screen instructions.
To upgrade from Ubuntu 9.04 on a server system: install the update-manager-core package if it is not already installed; edit /etc/update-manager/release-upgrades and set Prompt=normal; launch the upgrade tool with the command sudo do-release-upgrade; and follow the on-screen instructions. "

Upgrading Using the Alternate CD/DVD
Use this method if the system being upgraded is not connected to the Internet.
1.
Download the alternate installation CD
2.
Burn the ISO to a CD and insert it into the CD-ROM drive of the computer to be upgraded.

If the ISO file is on the computer to be upgraded, you could avoid wasting a CD by mounting the ISO as a drive with a command like:
sudo mount -o loop ~/Desktop/ubuntu-9.10-alternate-i386.iso /media/cdrom0
3. A dialog will be displayed offering you the opportunity to upgrade using that CD.

If the upgrade dialog is not displayed for any reason, you may also run the following command using Alt+F2:
gksu "sh /cdrom/cdromupgrade"
Or in Kubuntu run the following command using Alt+F2:

How did you manage to get yours working? When I'm asked to type in a mirror, there are only options for debian and when I tried http://ftp.is.co.za/debian, it kept saying 'file not found' for all the files.

For a fairly standard install, with very little extra software, you're looking at 1 to 2 gigs. This is because EVERYTHING on your system is upgraded. Even if your packages are up to date, they are only security and bug fixes, no new versions of software, which is why it's a full upgrade.
