Bootable Windows 10 on USB ... how?

As per title.

I'm using Rufus with a Windows 10 image file, but I can't boot up with it.

What haven't I done, or done wrong?

Have you checked BIOS/EFI settings? You may have to disable secure boot and select USB boot as yuor first preference.

i use rufus with win10 iso images all the time, what exactly is it doing or not doing?

If you have UEFI then no need to muck about with third party software...

1. Format usb to FAT32
2. Mount ISO and copy files to USB
3. Boot from said usb
4.
5. PROFIT!

Same here, never had any issues. Rufus is an amazing little utility.

Chances are the issue is not the usb once rufus is done with it, check maybe that the partition scheme is set to 'MBR partition scheme for BIOS or UEFI-CSM'
