I would minimize the 6 buttons, to maybe a P1 start, P2 start - and then two others, for pause and exit. I would also move them away to the corners or something
6 buttons is also enough for each player.
I would also label the extra buttons, but I don't know what they do.

I think it would be best to make it like this:
P1-start, "SHIFT Button", P2-start
P1 joy, 6 Buttons
P2 joy, 6 Buttons
the shift button at the top, works with EVERYTHING
Shift + Joy up/down = volume
Shift + P1 Start = Exit
Shift + P2 Start = Pause
etc,
etc,
easiest solution IMO.. you can even put a little name beneath some of the buttons, like "Exit" - which means if you hold the "shift" and press it, it will exit
a Second function for each button if you will...
You just need this:
[Shift]
[P1 Start]........[P2 Start]
(Joystick) [1]..[2]..[3]..............(Joystick) [1]..[2]..[3]..............
(Joystick) [4]..[5]..[6]..............(Joystick) [4]..[5]..[6]..............
If you hold in [shift], then [P1 Start] becomes Exit, and [P2 Start] becomes Pause
The P1 Joystick could control volume, and P2 Joystick speed/frameskip/whatever
then you have 12 other buttons to do whatever you need to do - all because you added 1 single little button - the shift button.
and yet, the controls remain SIMPLE - a Start button, joystick and 6 buttons for each player.....

Ok I didn't know you could do that in mame. On the Ultrastik, 8 is used as a shift button, so you could map 15 hardware buttons with just 8 on each joystick. So the way I made my CP is almost the same as yours, except that I have a shift button for each payer. So instead of 3 buttons on top, I have 4.
Why 4 buttons on top? Why not just use the 1-player and 2-player buttons as the shift button?

it would be like making your "A-key" on a keyboard to perform Shift..... a Shift button should have no other function except for shift.
Perspex: Maizey Plastics, there are a few of these around joburg - Even Builders Warehouse carries Perspex. s
Then make button 6 the shift key? And button 6 + 1-player for example = pause. There's lots of combinations that you will never press by accident.
